If you have auto sync on, then iTunes won't let you drag stuff on or off your iPod. iTunes does it all for you. Set it to manual and you can drag the entire library over and which ever playlists you want. Drag and drop or delete directly from the iPod using iTunes only when you've told iTunes you want to manage it. You won't get play count or ratings updated between the two without auto syncing, but some of people don't use that information.
Another way to get your results is to create a folder in iTunes and put as many playlists as you wish in it (include one that has all your music) and tell iTunes to sync to that folder.
